An automated trihydroxyindole procedure for the differential analysis of catecholamines.

چکیده
An automated trihydroxyindoleprocedure for the differential estimation of epinephrine and norepinephrinein tissues, urine, and blood is presented in detail. After an initial purification of the catecholaminesby adsorption on aluminum oxide, the neutralized eluates are subjected to automatic analysis, in which potassiumferricyanide and an alkaline stabilizing agent are automatically mixed sequentially with the dialyzed catecholamines. The fluorescent trihydroxyindole derivatives formed are then delivered to a flow cell in a fluorometer and the fluorescence recorded. A differential estimation is obtained by running samplesthrough the system, first with ascorbicacid stabilization to obtain the combined fluorescenceof both amines and then with thioglycolic acid stabilization to obtain the fluorescence of norepinephrinealone.

Semi-automated method for the differential determination of plasma catecholamines.
A method is described for the estimation of adrenaline and noradrenaline in a 5 to 10 ml plasma sample.A batch technique of adsorption and elution from alumina is used and the final estimation is carried out using an automated fluorimetric technique based on the trihydroxyindole method. Fluorescence is stabilized first by thioglycollic acid to give noradrenaline concentrations only and then by ...
متن کاملAutomated analysis for plasma epinephrine and norepinephrine by liquid chromatography, including a sample cleanup procedure.
We describe a fully automated method of analysis for plasma epinephrine and norepinephrine by use of a two-column system of "high-performance" liquid chromatography. Catecholamines in deproteinized plasma are purified on the first (preparation) column, then transferred automatically to the second (analytical) column in which epinephrine and norepinephrine are resolved. These compounds are then ...
متن کاملUse of constant temperature and sodium borohydride in the trihydroxyindole method for catecholamines.
Small temperature variations during fluorescent analysis with the trihydroxyindole method can introduce significant errors in the measurement of 15 p.g./L. or less of epinephrine or norepinephrine. To minimize temperature differences, we have devised a simple, constant-temperature system for the Farrand fluorometer. -Decomposition of ascorbic acid prior to and during analysis also must be preve...
متن کاملComparison of improved fluorometric methods used to quantitate plasma catecholamines.
Comparative studies reveal that the ethylenediamine and the trihydroxyindole method when employed as suggested are suitable for assessing sympatho-adrenal activation under various stressful conditions that are accompanied by release of catecholamines into the circulation. Both methods are very accurate in quantitating epinephrine or norepinephrine that has been added to plasma or whole blood, a...
متن کاملModified fluorometric method for determining plasma catecholamines.
Plasma catecholamines were fluorometrically determined by a modification of the standard alumina-trihydroxyindole method in which radioactive tracer catecholamines are used to follow the chromatographic behavior of the endogenous catecholamines on alumina. This modification permits appropriate fluorometry of that portion of the eluate with the highest concentration of catecholamines, because in...
